چغندرقند (Beta vulgaris L.) یکی از دو محصول اصلی و مهم تولید قند در دنیاست. بررسی عملکرد ژن­ها و همچنین معرفی
ژن­های ارزشمند، نیازمند استفاده از روش­های مهندسی ژنتیک است. از این­رو، استفاده از کشت بافت برای بهره­مندی از هر نوع روش انتقال ژنی گریزناپذیر است. بنابراین به­منظور تولید گیاهچه در گیاه سرسخت چغندر
­قند به­منظور یافتن ریز نمونه مناسب برای تولید جوانه به­روش باززایی مستقیم بدون نیاز به کشت مجدد بذرها و تهیه­ی جوانه انتهایی، از یک آزمایش فاکتوریل در قالب طرح کاملاً تصادفی استفاده شد. از جوانه­های نابجا موجود بر روی برگ­های حاوی پایه جوانه در روش باززایی مستقیم و از پهنک برگ و دمبرگ در دو موقعیت نزدیک و دور نسبت به جوانه رأسی دو لاین چغندرقند SBSI-04) و (SBSI-02 استفاده شد. پس از گذشت چهار هفته و دو بار واکشت، ریزنمونه­ها در محیط MS با ترکیب هورمونی IBA (1/0میلی­گرم در لیتر) و
 BAP (25/0 میلی­گرم در لیتر)، تعداد جوانه­های تولیدی شمارش و به­صورت درصد مورد مقایسه قرار گرفتند. نتایج این مطالعه نشان داد که ریزنمونه­ی دمبرگ بیشترین جوانه را تولید نمود. همچنین از نظر صفت درصد جوانه­زایی، لاین SBSI-04 در مجموع نسبت به لاین SBSI-02 جوانه­های بیشتری را تولید کرد. به­طور­کلی، ریز نمونه­های دمبرگ داخلی لاین SBSI-04 بیشترین قابلیت تولید جوانه را نشان دادند، این موضوع نشان داد که برای تولید گیاهان تراریخت، ضمن توجه به اهمیت نوع ژنوتیپ بهتر است از کشت بافت این ریزنمونه و تراریزش آن استفاده شود.
