چکیده مبسوط
مقدمه و هدف: گوجه ­فرنگی (Solanum lycopersicum L.) یکی از مهم­ترین گیاهان زراعی متعلق به خانواده  Solanaceae می ­باشد. گوشتی بودن میوه از صفات مهم موثر در کیفیت میوه گوجه ­فرنگی بوده و توارث کمی دارد. از جمله ژن­های دخیل در گوشتی بودن میوه گوجه فرنگی ژن­های پکتین استراز 1 (PE1) و پلی گالاکتوروناز (PG) می­ باشد. با توجه به نقش مهم این ژن­ها در کیفیت میوه گوجه فرنگی، شناسایی چندشکلی­ های تک نوکلئوتیدی (SNPs) در مناطق کد کننده این ژن­ها برای تولید نشانگرهای عملکردی مرتبط با سفتی میوه ضروری می باشد.
مواد و روش ­ها: در این مطالعه 96 ژنوتیپ از 12 جمعیت جمع آوری شده از مناطق مختلف استان آذربایجان غربی (ایران) و ترکیه در گلخانه تحقیقاتی دانشکده کشاورزی دانشگاه ارومیه کشت شد. برای شناسایی SNPها در ژن‌های PE1 و PG، آغازگرهای اختصاصی با استفاده از نرم‌افزار FastPCR برای تکثیر قطعاتی از مناطق کدکننده این ژن‌ها در 96 ژنوتیپ گوجه‌فرنگی طراحی شد. برای هضم قطعات تکثیر شده دو ژن از آنزیم ­های برشی Pst1 و TruI استفاده شد. با توجه به عدم وجود چندشکلی در الگوهای برشی حاصل از هضم آنزیمی، چهار فرد از جمعیت­های مختلف انتخاب و قطعه تکثیری در این افراد خالص سازی و  توالی یابی شد. بعد از بازیابی قطعه تکثیری هر ژن، شناساییSNPها با استفاده از همردیفی توالی‌های هر ژن با نرم‌افزار Clustal Omega انجام گرفت.
یافته ­ها: بر اساس نتایج همردیفی، در ژن PE1 چهار SNP شناسایی شد که 75 درصد آنها از نوع همجنس با فراوانی 50 درصد A/G، 25 درصد  T/C و 25 درصد جهش­ها از نوع نا­همجنس  C/A بود. در ژن PG شش SNP شناسایی شد که 66/7 درصد آنها از نوع همجنس با فراوانی 33/3 درصد A/G، 3/33 درصد  T/C و 3/32 درصد جهش ­ها از نوع نا­همجنس با فراوانی 16/16 درصد T/A و 16/16 درصد G/C  بود.
نتیجه‌گیری: به طور کلی نتایج مطالعه حاضر نشان داد که میانگین تعداد SNPها در هر 100 جفت باز از اگزون‌ ژن‌های PE1 و PG به ترتیب 1/22 و 0/41 است. همچنین فراوانیSNP ها در ژن PG کمتر از PE1 بود. تعداد کم SNP های مشاهده شده در اگزون های هر دو ژن نشانگر حفاظت شدگی مناطق کد کننده این ژن ها در طول تکامل گوجه فرنگی می ­باشد. همچنین SNP های شناسایی شده در مطالعه حاضر می ­تواند در برنامه­ های اصلاحی گوجه فرنگی برای معرفی نشانگرهای عملکردی مرتبط با سفتی میوه مورد استفاده قرار گیرد.
