چکیده مبسوط
مقدمه و هدف: عارضه اختلال در غلاف‌بندی نوع ویژه­ای از رشد همراه با ناهنجاری در تشکیل گل و غلاف است که در آن گیاهان سویا مدت­ها پس از رسیدگی غلاف ­ها، سبز باقی می­ مانند و منجر به کاهش شدید عملکرد می‌گردد. به منظور بررسی و شناسایی پروتئین‌های دخیل در تظاهر عارضه اختلال در غلاف‌بندی سویا، پروفایل بیان پروتئین ­های گیاهان سالم و مبتلا به عارضه اختلال در غلاف‌بندی سویا در دو رقم کتول و گرگان 3 بر اساس رهیافت‌های پروتئومیک و بیوانفورماتیک مورد مقایسه قرار گرفتند.
مواد و روش‌ها: برای این منظور نمونه­ های برگی گیاهان سالم و مبتلا از دو رقم فوق در مرحله گلدهی انتخاب و پروتئین­ آنها به روش TCA/Acetone استخراج شد. سپس تفکیک پروتئین ­ها و تجزیه افتراقی آنها به ترتیب با استفاده از ژل الکتروفورز دو بُعدی و طیف‌سنجی جرمی nESI-LC-MS/MS انجام شد.
یافته‌ها: تجزیه و تحلیل نتایج ژل الکتروفورز دو بُعدی نشان داد که سطح بیان 5 تا از 155 لکه پروتئینی در رقم کتول و 11 تا از 143 لکه پروتئینی در رقم گرگان 3 در گیاهان سالم و مبتلا به طور معنی­داری تغییر یافت. آنالیز افتراقی پروتئین­ ها با تغییر بیان معنی ­دار باnESI-LC-MS/MS  نشان داد که در پاسخ به این عارضه بیشتر پروتئین­ های درگیر در فعالیت ­های تنظیم سلولی، تولید انرژی، متابولیسم، انتقال سیگنال، نسخه­ برداری و ترجمه ژن و نیز تقسیط و ذخیره‌سازی پروتئین­ها شرکت دارند.
نتیجه‌گیری: در میان پروتئین­ های شناسایی شده احتمالا پروتئین­های 14-3-3 like protein، OEE2 ، زیر واحد بزرگ رابیسکو و پروتئین NACA ممکن است از جمله تنظیم کننده­ های کلیدی عارضه اختلال در غلاف‌بندی سویا باشند.
