کاهو از خانواده کاسنیان، برگی، سرمادوست یک‌ساله، غنی از مواد معدنی، ویتامین­ ها و مواد مغذی است. هدف تحقیق حاضر ارزیابی تنوع ژنتیکی 15 رقم و جمعیت کاهو با استفاده از ناحیه ITS بود. کیفیت توالی‌ها با استفاده از نرم‌افزار Chromas 2.1.1 بررسی و سپس با روش ClustalW توسط نرم‌افزار MegAlign 5 هم‌ردیف سازی شده و دندروگرام روابط تبار­زایی و ماتریس تفاوت و تشابه توالی‌ها ترسیم گردیدند. همچنین مقایسه ­ای بین کاهوهای مورد استفاده در این تحقیق و سایر گیاهان جنس‌های مختلف خانواده کاسنیان (ارائه‌شده در سایت NCBI) صورت گرفت. مقدار عددی نسبت (dNdS) برابر 015/0 بود که نشان‌دهنده انتخاب خالص بر ژن مورد بررسی بوده و باعث تغییرات کلیدی نشده است. تجزیه خوشه‌ای، ژنوتیپ­ های مختلف کاهو را در 3 گروه تقسیم و نتوانست بر اساس موقعیت جغرافیایی از هم تفکیک کند؛ اما توانست کاهوهای این تحقیق و سایر گیاهان جنس‌های مختلف خانواده کاسنیان را از هم تفکیک کند. لذا می‌توان نتیجه گرفت که ITS می‌تواند ابزاری مناسب جهت ارزیابی‌های ژنتیکی بین‌گونه‌ای و بین جنسی باشد. کمتر از یک بودن عدد dNdS و تعداد کم جایگاه‌های حذف و اضافه (از 1551 جایگاه، 376) نشان‌دهنده تغییرات کم بین لاین‌های مختلف بوده لذا شاید عدم توانایی ITS در تفکیک درون گونه‌ای جمعیت‌ها به همین دلیل باشد. با توجه به اینکه منشأ یا خواستگاه اولیه گیاهان متعلق به مراکزی است که بیشترین تنوع را دارند و چون جمعیت­ های کاهو ورزنه اصفهان دارای بیشترین تنوع بودند در نتیجه ضرورت دارد در جمع­ آوری ژرم­ پلاسم ­های کاهو جهت بهره‌برداری‌های اصلاح نژادی به منطقه ورزنه اصفهان توجه بیشتری شود.
