چکیده مبسوط
مقدمه و هدف: توتون (Nicotiana tabacum L.) با 2n = 4x = 48 کروموزوم یکی از مهم ‏ترین گیاهان صنعتی و اقتصادی در بسیاری از کشورهای دنیا است که عمدتاً به‎ خاطر برداشت و جمع‎آوری برگ‎های آن کشت می‎شود. عملکرد توتون به ‎دلیل طبیعت پلی‎ژنیکی که دارد، شدیداً تحت تاثیر تنش‎های غیر زنده و زنده از جمله علف ‏هرز گل‏ جالیز قرار می‎گیرد. در این میان، واکنش واریته‎ها به محیط بسته به ژنوتیپ رقم متفاوت است که از آن به برهمکنش ژنوتیپ × محیط یاد می‎شود. با بررسی برهمکنش ژنوتیپ در محیط و تجزیه پایداری می‎توان ژنوتیپ‌هایی را انتخاب نمود که در انواع مختلف محیط‎ها از عملکرد مطلوبی برخوردار باشند. اگرچه در مورد استفاده از روش‎های پایداری در ارزیابی واکنش گیاهان به محیط گزارشات زیادی وجود دارد، ولی در مورد پایداری عملکرد برگ خشک توتون در محیط‎ های آلوده به گل‎ جالیز گزارشی وجود ندارد. این پروژه با هدف ارزیابی واکنش ژنوتیپ‎های توتون شرقی موجود در ژرم‎پلاسم توتون کشور در دو محیط گل‏ جالیزدار و بدون گل‏ جالیز به‎ منظور شناسایی و انتخاب ژنوتیپ‎ های سازگار و پایدار در محیط‎ های مورد مطالعه انجام گردیده است.
مواد و روش‎ ها: در این آزمایش، 92 ژنوتیپ توتون تیپ شرقی و نیمه ­شرقی از لحاظ تحمل به تنش گل ­جالیز در طی دو سال زراعی در قالب طرح بلوک‎ های کامل تصادفی با سه تکرار مورد ارزیابی قرار گرفتند. ژنوتیپ‎های توتون از بانک ذخایر ژنتیکی و زیستی مرکز تحقیقات توتون ارومیه و انستیتو تحقیقات توتون تیرتاش (مازندران) تهیه شدند. برای کشت، گلدان‎های سفالی به حجم 10 لیتر انتخاب و با خاکی که از مزرعه یونجه تهیه شده بود، پُر شدند و گلدان‎ها بر اساس نقشه طرح در فضای باز در محوطه مرکز تحقیقات توتون ارومیه چیده شدند. یونجه میزبان گل جالیز نیست و استقرار طولانی مدت آن در مزرعه (معمولاً شش سال) باعث می‎شود هرگونه بذر گل جالیز موجود در خاک از بین برود؛ بنا بر این، خاک تقریباً عاری از گل جالیز شود. در محیط گل‎ جالیزدار، خاک گلدان‎ها قبل از پرشدن با 0/06 گرم بذر گل‏ جالیز گونه Orobanche cernua (حاوی تقریباً 12000 بذر) که از مزرعه مرکز تحقیقات توتون ارومیه در سال قبل از اجرای آزمایش جمع‎آوری شده بود، مخلوط شد. در مناطق شمال غرب و مخصوصاً در آذربایجان غربی، این گونه گل ‎جالیز روی گیاه توتون غالب است و در مزارع بیشتر دیده می‎شود. نشاء هر یک از ژنوتیپ‎های توتون در خزانه تهیه شد و وقتی گیاهچه‎های توتون به ارتفاع 12 سانتی‎متر رسیدند، به گلدان‎ها منتقل شدند. برگ‎های ژنوتیپ‎های توتون در زمان رسیدگی صنعتی (حدود 45 تا 50 روز بعد از نشاکاری) در سه نوبت (پابرگ، کَمَر برگ و لَچِه برگ) برداشت، در آفتاب خشک، و با استفاده از ترازوی دیجیتال با دقت 0/001 گرم وزن شدند. پس از انجام تجزیه واریانس ساده و تجزیه واریانس مرکب، به‎ علت وجود اثر متقابل ژنوتیپ × محیط آماره‎های تک ­متغیره پایداری محاسبه شدند.
یافته‎ها: نتایج تجزیه واریانس نشان دادند که بین ژنوتیپ‎های توتون از نظر تمامی صفات مورد مطالعه اختلاف معنی‎دار وجود داشت و اثر متقابل ژنوتیپ × محیط در مورد تمامی صفات معنی‎دار بود که حاکی از وجود تنوع بین ژنوتیپ‎ها و عکس‎العمل متفاوت آن‎ها به شرایط عدم تنش و تنش گل جالیز است. مقایسات میانگین نشان دادند که تنش گل جالیز میانگین کلیه صفات مورفولوژیک را کاهش می‎دهد. براساس نتایج روش‎های تجزیه پایداری مبتنی بر تجزیه واریانس و تجزیه رگرسیون، ژنوتیپ‎های شماره 45، 33 و 78 به‎ ترتیب دارای بیشترین مقدار ضریب تغییرات و به‎ عبارتی کمترین میزان پایداری بودند؛ در این بین، ژنوتیپ 45 میانگین بالاتر از میانگین کل داشت. ژنوتیپ‎های شماره 75، 53 و 41 به علت داشتن کمترین مقدار واریانس پایداری، جزء ژنوتیپ‎های پایدار بودند. بر اساس نتایج روش پلستید و پترسون (θi)، ژنوتیپ‎های شماره 75، 55، 53 و 41 کمترین مقدار θi را داشتند و در بین آن‎ها ژنوتیپ‎ 53 عملکردی بالاتر از میانگین کل داشت.
نتیجه‎گیری: معرفی و استفاده از شاخص‎های پایداری، به‎ واسطه ارزیابی پایداری مواد ژنتیکی تا حد زیادی می‎تواند در گزینش هدفمند ژنوتیپ‎ها در راستای انتخاب ژنوتیپ‎های مطلوب و کارآمد و توسعه ارقام پُرمحصول و متحمل به تنش سودمند باشد. بر اساس روش‎های مختلف پایداری، ژنوتیپ‎ شماره 53 به‎ عنوان ژنوتیپ‎ پایدار با عملکرد مطلوب معرفی می‎شود.
