چکیده مبسوط
مقدمه و هدف: بیان ژن‎ها تحت تاثیر مراحل مختلف رشدی و عوامل مختلف محیطی متغییر است. تنش خشکی در مرحله گل‎دهی و پرشدن دانه که به‎عنوان تنش خشکی پایان فصل شناخته می‎شود، می‎تواند منجر به کاهش شدید عملکرد یا شکست کامل تولید محصول شود. تجزیه و تحلیل ژنتیکی مقاومت به خشکی در مرحله زایشی به‎منظور درک مکانیسم واکنش گیاه به شرایط خشکی در مواجهه با چالش‎های حفظ امنیت غذایی ضروری است. بررسی پروفایل رونوشت ژن‎ها در بافت‎ها و مراحل مختلف رشدی تحت شرایط مختلف تنش‎های محیطی می‎تواند بینشی در مورد مکانیسم‎های مولکولی و نحوه‎ی واکنش گیاهان به تنش ارائه دهد. جو به‎عنوان یک گیاه مدل برای رمزگشایی مکانیسم‎های تحمل به خشکی شناخته شده است و مطالعه مکانیسم‎های مولکولی جو برای اصلاح محصولات مهم است، زیرا توانایی تحمل محدودیت‎های آبی در مراحل گلدهی و پر شدن دانه را دارد. هدف این پژوهش شناسایی ژن‎های دارای بیان افتراقی تحت تنش خشکی پایان فصل در گیاه جو با استفاده از تکنیک RNA-Seq بود. براساس مطالعه‎ی امینی و همکاران روی 13 ژنوتیپ جو دو ردیفه‎ی بهاره تحت تنش خشکی، ژنوتیپ Dayton/ Ranney (اصلاح شده توسط ICARDA) به‎عنوان یک ژنوتیپ متحمل به خشکی شناسایی شد و در این مطالعه برای بررسی پروفایل بیان ژن گیاه جو تحت تنش خشکی انتهای فصل استفاده شد.
مواد و روش‎ها: ژنوتیپ جو بهاره Dayton/ Ranney در مرحله ظهور برگ پرچم در معرض تیمار تنش خشکی 70 درصد تخلیه آب در دسترس قرار گرفت. RNA کل از برگ‎های گیاهان شاهد و تنش دیده استخراج و کیفیت RNA استخراج شده بررسی شد. پس از توالی‎یابی، آنالیز انجام شد و الگوی بیان ژن‎های دارای بیان افتراقی تحت تنش خشکی پایان فصل به‎دست آمد. همچنین، ژن‌های دارای بیان افتراقی، از نظر عملکردی با استفاده از آنالیز غنی‌سازی ژن بررسی شدند. محل اتصال عوامل رونویسی در توالی راه‎انداز ژن‎های دارای بیان افتراقی با استفاده نرم‎افزار آنلاین PlantPAN 3.0 شناسایی و فراوانی جایگاه‎های اتصال به‎صورت درصدی از کل جایگاه‎های شناسایی شده گزارش شد.
یافته‎ها: تحت تنش خشکی انتهای فصل، در گیاه جو 2920 ژن افزایش بیان و 2289 ژن کاهش بیان معنی‎دار نشان دادند. ژن‎های شناسایی شده در فرایندهای فتوسنتز، متابولیسم کربوهیدرات و لیپیدها، فرایندهای تنظیمی، پاسخ به محرک‎های غیر‎زیستی و تنش، نمو و بلوغ بذر دخیل بودند. همچنین، براساس آنالیز هستی‎شناسی ژن، این ژن‎ها در فرایندهای متابولیکی و بیوسنتزی اسید کربوکسیلیک، متابولیسم ساکارز و گلوکان سلولی، پروتئولیز، فسفوریلاسیون، متابولیسم و بیوسنتز RNA و متابولیسم اسیدآمینه خانواده سرین نقش داشتند. از ژ‎ن‎های با بالاترین افزایش بیان تحت تنش خشکی می‎توان به خانواده پروتئین‌های تجمعی در اواخر جنین‎زایی، یک ژن مسیر فنیل پروپانوئید به‎نام آنترانیلات-N-بنزوئیل ترانسفراز 1، ژن زایلوگلوکان اندوترانس گلوکوزیلات/هیدرولاز، پروتئین سرین/ترئونین فسفاتاز، یک ناقل میتوکندریایی آرژینین، یک ژن اندونوکلئاز، آنزیم لاکاز و چندین عامل رونویسی اشاره نمود. همچنین ژن‎هایی که بیشترین کاهش بیان معنی‎داری را تحت تنش خشکی نشان دادند شامل یک کیناز گیرنده حاوی دامنه لکتین نوع L (Hv-LecRK)، یک ژن شبه-ریبونوکلئاز 3، عامل رونویسی HEC1-like، لیپواکسیژناز القا‎‎شونده با متیل جاسمونات II، گلوکان اندو-3،1-بتا-گلوکوزیداز GIII، آکواپورین PIP2-5، پروتئین 70 کیلو دالتونی شوک حرارتی و ژن آسپارتیک پروتئیناز نپنتزین-1 بود. همچنین، دو ژن ناشناخته 2HG0195510 و 4HG0389440 افزایش بیان قابل ملاحظه‎ای را نشان دادند. این ژن‎ها در فرایندهای متابولیکی و بیوسنتزی اسید کربوکسیلیک، پاسخ به محرک‎های غیر‎زیستی و تنش، پاسخ به محرک‎های درون‎زا، پروتئولیز، فسفوریلاسیون، متابولیسم و بیوسنتز RNA، فرایند متابولیک پروتئین، متابولیسم اسیدآمینه خانواده سرین و حمل و نقل نقش داشتند. همچنین در ﺳﻄﺢ عملکرد مولکولی، برای کلیه ژن‎های دارای بیان افتراقی، گروهﻫﺎی ﻓﻌﺎﻟﯿﺖ کاتالیزوری و اﺗﺼﺎل، ﺑﯿﺸﺘﺮیﻦ ﺗﻌﺪاد ژن را ﺑﻪ‎ﺧﻮد اﺧﺘﺼﺎص دادﻧد. سایر عملکرد‎های مولکولی شناسایی شده برای ژن‎های پاسخگو به تنش خشکی شامل اتصال به پروتئین، اتصال به نوکلئوتیدی، فعالیت حمل و نقل، اتصال به DNA، فعالیت ترانسفراز، کیناز، هیدرولاز و پیروفسفاتاز می‎باشد. علاوه براین، ژن‎های افزایش بیان یافته به‎طور خاص دارای عملکردهای انتقال پیام، عامل رونویسی، تنظیم آنزیم، انتقال مولکولی و فعالیت گیرنده بودند. جایگاه‎های اتصال عوامل رونویسی شناسایی شده در ژن‎های دارای بیان افتراقی به 64 خانواده طبقه‎بندی شدند. بیشترین درصد جایگاه‎های اتصال در ژن‎های افزایش بیان یافته متعلق به عوامل رونویسی ERF/AP2 و پس از آن فراوان‎ترین جایگاه‎های اتصال متعلق به خانواده‎ها‎ی عوامل رونویسی bZIP، bHLH، DOF و GATA بود. همچنین فراوان‎ترین جایگاه‎های اتصال در ژن‎های کاهش بیان‎یافته شامل AP2/ERF، BES1 ، EIL، TCP، Myb/SANT، GATA و DOF بود.
نتیجه‎گیری: با بررسی بیان ژن تحت تنش خشکی پایان فصل جنبه‎هایی از مکانیسم مقاومت گیاه جو به تنش خشکی که با فعالیت‎های متابولیکی و بیوسنتزی گیاه در مرحله زایشی مرتبط است شناسایی شد. نتایج نشان داد که شبکه‎های ژنی متنوع و پیچیده‎ای در پاسخ گیاه جو به تنش خشکی انتهای فصل نقش داشتند و عمدتاً فرایندهای زیستی مرتبط با فتوسنتز و تولید متابولیت‎های پیش‎ساز و انرژی کاهش و فرایند‎های متابولیکی افزایش یافت. همچنین فرایند پاسخ به محرک در هر دو مجموعه از ژن‎های افزایش و کاهش بیان یافته، مشاهده شد.
