چکیده مبسوط
مقدمه و هدف:
کاپاریس اسپینوزا متعلق به جنس Capparis و خانواده Capparidaceae دارای 350 گونه است. این گیاه با نام تجاری کَبَر دارای گونه­ های متعددی در ایران است. کبر دارای پتانسیل­های عظیمی برای کشاورزی و بهره ­برداری، به دلیل بازار بین ­المللی متنوع نشان می­دهد. امروزه تمرکز بر انتخاب و اصلاح این گونه در بسیاری از کشورها انجام شده ‌است. انواع تکنیک‌های تحلیلی مختلف ژنتیکی در زمینه ژنتیک مولکولی به همراه چندین نشانگر ژنتیکی مبتنی بر PCR برای ارائه اطلاعات تغییرات ژنتیکی در گونه‌های گیاهی استفاده می‌شوند. نشانگرهای ISSR و SCoT نشانگرهای غالبی هستند که می ­توانند برای ارزیابی تنوع ژنتیکی در ژرم­پلاسم گیاهان استفاده شوند.
مواد و روش‌ها: برای ارزیابی ساختار جمعیت Capparis spinosa که به طور طبیعی در مناطق غرب ایران رشد می‌کنند، 80 ژنوتیپ از 12 مکان مختلف جمع‌آوری شد.
یافته‌ها: خوشه‌بندی با استفاده از دو نشانگر ISSR و SCoT، 80 ژنوتیپ را در دو گروه توزیع کرد. طبق نتایج تجزیه ارتباط صفات مورفولوژیکی بر اساس دو نشانگر ISSR و SCoT، همه نشانگرهای گزارش شده معنی­ دار بودند. در مجموع بر اساس نتایج، 966 نشانگر SCoT و 571 نشانگر ISSR که ارتباط معنی­ داری (در سطح احتمال 5 درصد) با صفات مورد مطالعه داشتند شناسایی شدند.
نتیجه‌گیری: طبق گروه­بندی استراکچر با استفاده از نشانگر SCoT، نمونه­ های مربوط به جمعیت­ های جمع­ آوری شده از کرمانشاه، چارمله، ایلام، سرپل­ذهاب، قصرشیرین، گورسفید، گیلان، خسروی، نفت شهر، و سومار در یک گروه و نمونه­ های مربوط به جمعیت­ های جمع­ آوری شده از کرند و ایوان در یک گروه طبقه­ بندی شدند. طبق گروه ­بندی استراکچر با استفاده از نشانگر ISSR، نمونه ­های مربوط به جمعیت­ های جمع­ آوری شده از کرند، خسروی، نفت شهر، و بیشتر نمونه­ های جمع­آوری شده از سومار در یک گروه، و نمونه­ های مربوط به جمعیت­ های جمع­ آوری شده از کرمانشاه، چارمله، ایلام، ایوان، سرپل­ذهاب، قصرشیرین، گورسفید، گیلان و بخشی از نمونه­ های سومار در یک گروه طبقه ­بندی شدند. به طور کلی نشانگرهای ISSR و SCoT تنوع ژنتیکی بالایی در نمونه­ های جمع ­آوری شده از غرب ایران را نشان دادند، و نتایج تجزیه ارتباط نشان داد که استفاده از روش GLM همراه با ترکیبی از نشانگرهای مختلف می ­تواند در شناسایی نشانگرهای مرتبط با صفات مورفولوژیک در کبر مؤثر باشد.
