fa استفاده از نشانگرهای آلل اختصاصی در شناسایی آلل های مکان های ژنی VRN-1 در ژنوتیپ‌های گندم نان Application of Allele Specific Markers in Determining Alleles at VRN-1 Loci in Bread Wheat Genotypes اصلاح نباتات مولكولي اصلاح نباتات مولكولي پژوهشي Research <div style="text-align: justify;">     گندم مهمترین گیاه زراعی است و در سرتاسر جهان کشت می شود و سازگاری گسترده ای با مناطق گوناگون اقلیمی دارد. یکی از عوامل مهمی که باعث سازگاری گندم می‌شود، ژن های بهاره سازی است. در برنامه‌های به نژادی گندم شناخت از نوع آلل‌ها در مکان‌های ژنی کنترل کننده بهاره‌سازی می‌تواند نقش مهمی در معرفی ارقام سازگار به مناطق مختلف داشته باشد. در سطح مولکولی سه مکان ژنی برای کنترل بهاره سازی در گندم نان وجود دارد. این مکان های ژنی Vrn-1،  Vrn-2و  Vrn-3هستند. نیاز بهاره سازی در گندم هگزاپلویید عمدتاً توسط مکان ژنی Vrn-1تعیین می شود که شامل مکان ژنی Vrn-A1، Vrn-B1 و Vrn-D1 است. مکان های ژنی Vrn-A1، Vrn-B1 و Vrn-D1 برای عادت رشد بهاره غالب و اپیستاتیک بر آلل‌های عادت رشد زمستانه هستند، بنابراین ارقام زمستانه دارای آلل‌های مغلوب در سه مکان ژنی VRN-1 است. در این مطالعه 34 ژنوتیپ گندم نان خاورمیانه به همراه ژنوتیپ گندم بهاره چینی با استفاده از نشانگرهای آلل اختصاصی برای مطالعه تنوع آللی در مکان ژنی Vrn-1 مورد بررسی قرار گرفتند. مطالعه حاضر نشان داد که در مکان ژنی VRN-A1 پنج ژنوتیب دارای آلل غالب VRN-A1a بود.آلل VRN-A1b در هیچ یک از نمونه ها مشاهده نشد. تنها یکی از نمونه ها آلل VRN-A1c را نشان داد و 32 ژنوتیپ به همراه گندم بهاره چینی دارای آلل مغلوب vrn-A1 بودند. یک ژنوتیپ با شماره شش در این مکان ژنی بیش از یک آلل را نشان داد که غیرمعمول بود. در مکان ژنی VRN-B1، 14 ژنوتیپ دارای آلل غالب و 20 ژنوتیپ به همراه ژنوتیپ بهاره چینی دارای آلل مغلوب بودند.</div> <div style="text-align: justify;">    Wheat is the most important crop and is cultivated all over the world. Wheat has a wide range of adaptability to different climates. Vernalization genes are one of the important factors determining wheat adaptability. in wheat breeding program understanding the alleles at vernalization requirement gens are useful to introduce new cultivar for different climates. At the molecular level, the length of the vernalization period of common wheat (Triticum aestivum L.) is determined mainly by three loci: VRN-1, VRN-2 and VRN-3. The VRNA1, VRN-B1, and VRN-D1 genes are dominant for spring growth habit and epistatic to the alleles for winter growth habit. Therefore, winter cultivars are homozygous for the recessive alleles at the three VRN-1 loci. In the current study, 34 bread wheat accessions plus Chinese Spring wheat were investigated for allelic variation at Vrn1 loci. The current study showed that five genotypes had dominant allele at VRN-A1. VRN-A1b allele was not found at any of the genotypes. Only one genotype had VRN-A1c allele and 32 genotypes including the Chinese Spring showed vrn-A1 allele. The genotype No. 6 unexpectedly had more than one allele. At locus VRN-B, 14 genotypes had dominant allele while 20 including the Chinese Spring showed recessive allele. ele was not found at any of the genotypes.
