fa ارزیابی مقاومت گیاهچه‌ای ارقام تجاری گندم نان نسبت به بیماری زنگ سیاه با استفاده از روش GGE بای‌پلات Evaluation of Seedling Stage Resistance of Commercial Bread Wheat Cultivars to Black Rust Disease using GGE Biplot Method اصلاح نباتات General پژوهشي Research <div style="text-align: justify;">چکیده مبسوط مقدمه و هدف: گندم در تغذیه جامعه بشری به‌ویژه در کشورهای جهان سوم دارای اهمیت ویژه‌ای است. زنگ ساقه یا زنگ سیاه شایع‌ترین، گسترده‌ترین و ویرانگرترین بیماری‌ مزارع گندم در ایران و سراسر جهان به شمار می‌رود. دامنه خسارت این بیماری با توجه به‌شدت آن می‌تواند از کاهش عملکرد تا نابودی کامل مزارع متغیر باشد. توسعه ژنوتیپ‌های دارای ژن مقاومت به بیماری زنگ سیاه یکی از راهکارهای مهم برای مقابله با این بیماری مخرب محسوب می‌شود. شناسایی منابع مقاومت و وارد نمودن آن‌ها در برنامه‌های به‌نژادی موجب تولید ژنوتیپ‌های مقاوم می‌گردد. مواد و روش‌ها: تعداد 89 رقم تجاری گندم نان به همراه شاهد حساس موراکو با هدف شناسایی ارقام مقاوم به زنگ سیاه در مرحله گیاهچه‌ای نسبت به شش نژاد قارچ عامل بیماری در قالب طرح بلوک‌های کامل تصادفی با دو تکرار تحت مطالعه قرار گرفت. کلیه مراحل ارزیابی گیاهچه‌ای در گلخانه‌های تحقیقاتی واحد پاتولوژی بخش تحقیقات غلات مؤسسه تحقیقات اصلاح و تهیه نهال و بذر کرج طی سال 1398 انجام شد. در مدل گرافیکی GGE biplot هر یک از نژاد‌های بیماری (PKTTF، PKSTC، PKSTF، PTRTF، TTTTF و PTTTF) به‌عنوان یک محیط تلقی گردیدند و تجزیه گرافیکی بر اساس مقادیر منفرد پاسخ ارقام نسبت به نژادها انجام شد. یافته‌ها: نژادهای PKTTF، PKSTC، PKSTF، PTRTF و TTTTF بیشترین میزان همبستگی را روی نمودار داشتند، به همین دلیل یک گروه بزرگ را به وجود آوردند؛ ارقام پیشتاز، سیوند، عدل، پیشگام، نیک‌نژاد و الوند در برابر نژادهای این گروه دارای واکنش مقاومت بالایی بودند. نژاد PTTTF به‌تنهایی گروه دوم را تشکیل داد و ارقام هامون، مرودشت، زرین، گهر، داراب 2 و سبلان در برابر این نژاد از واکنش مقاومت بالایی برخوردار بودند. نمودار بای‌پلات رتبه‌بندی ارقام بر اساس میانگین تیپ آلودگی و پایداری حاکی از این بود که ارقام پیشتاز و عدل دارای واکنش مقاومت پایدار نسبت به هر شش نژاد بیماری می‌باشند، نتایج این نمودار توسط نمودار رتبه‌بندی ارقام از نظر مقاومت بر طبق رقم ایده‌آل فرضی نیز تأیید شد. رتبه‌بندی شدت بیماری‌زایی نژادهای تحت بررسی بر اساس نژاد فرضی، مؤید بیماری‌زایی بالای نژاد PTTTF بر روی ارقام تحت مطالعه بود. ترتیب بیماری‌زایی سایر نژادها از بیشترین به کم‌ترین عبارت از PKTTF، TTTTF، PTRTF، PKSTF و PKSTC بود. نتیجه‌گیری: روش چندمتغیره گرافیکی GGE بای‌پلات در بررسی مقاومت ارقام تجاری گندم مؤثر بود و اطلاعات ارزشمندی را در خصوص شناسایی ارقام برخوردار از ژن‌های مقاومت به بیماری زنگ سیاه ارائه کرد که با وارد نمودن این ارقام به برنامه‌های به‌نژادی، می‌توان گام مؤثری در جهت توسعه ارقام حائز مقاومت در برابر این بیماری طی نمود و به مقابله ژنتیکی با آن پرداخت.</div> <div style="text-align: justify;">Extended Abstract Introduction and Objective: Wheat is of great importance in the nutrition of human society, especially in third world countries. Stem rust or black rust is the most common, widespread and destructive wheat disease in Iran and around the world. The extent of damage of this disease, depending on its severity, can vary from reduced yields to complete destruction of farms. The development of genotypes with the black rust resistance gene is one of the most important strategies to combat this destructive disease, considering the importance of wheat in the nutrition of human society, especially in third world countries. Identifying sources of resistance to import them in breeding programs leads to the development of resistant genotypes. Material and Methods: 89 bread wheat cultivars along with Morocco as susceptible control were studied with the aim of identifying stem rust resistance cultivars in the seedling stage to six races of the disease in randomized complete block design with two replications. All stages of plant assessment were carried out in the Cereal Research Department, Seed and Plant Improvement Institute (SPII), Karaj, Alborz, Iran, in 2019. In the GGE Biplot graphics model, each disease races (PKTTF, PKSTC, PKSTF, PTRTF, TTTTF and PTTTF) were considered as an environment, Therefore, based on the singular value decomposition of cultivars' response to races, graphical analysis was performed. Results: PKTTF, PKSTC, PKSTF, PTRTF and TTTTF were classified into a group based on the degree of correlation between them and Pishtaz, Sivand, Adl, Pishgam, Niknezhad and Alvand cultivars had resistance to those races. PTTTF race was placed in the second group and Hamoon, Marvdasht, Zarrin, Gohar, Darab 2 and Sabalan cultivars showed resistance against this race. Pishtaz and Adl illustrated stable resistance reaction for all six studied races. The severity of virulence of the studied races confirmed the high pathogenicity of PTTTF on the investigated germplasm. The orders of pathogenicity of other races from highest to lowest were PKTTF, TTTTF, PTRTF, PKSTF and PKSTC. Conclusion: The multivariate graphical GGE biplot method was effective in investigating the resistance of commercial wheat cultivars and provided valuable information on the identification of cultivars with genes resistant to black rust disease.
