امواج فراصوت دارای کابردهای متعددی در صنعت، پزشکی و فناوری‌ زیستی است. امواج فراصوت کم انرژی، نفوذ‌پذیری غشای سلولی را افزایش داده و منجر به اثرات بیولوژیکی متعددی در سلول‌های گیاهی می‌شود. این تحقیق با هدف بررسی تأثیر تیمار امواج فراصوت بر زنده­مانی و رشد ریزنمونه­های نوک ساقه گیاه اسپرس انجام گرفت. برای این منظور، ریز نمونه­های نوک ساقه تحت تأثیر تیمار امواج فراصوت (فرکانس 37 کیلوهرتز) با زمان‌های صفر (شاهد)، 20، 30، 40، 50، 60، 90، 180،120، 240 و 300 ثانیه در حمام اولتراسونیک قرار گرفته و سپس روی محیط کشت MS حاوی 1/0 میلی­گرم در لیتر IBA و 3 میلی­گرم در لیتر BAP کشت گردیدند. نتایج نشان داد که درصد ساقه‌دهی ریزنمونه‌ها به‌طور معنی‌داری در اثر تیمار امواج فراصوت کاهش یافته ولی درصد ساقه‌های چندگانه، تعداد ساقه در هر ریزنمونه و درصد کالوس‌زایی به طور معنی‌داری افزایش یافته است. تیمار شاهد (بدون امواج فراصوت) بیشترین درصد ساقه­دهی به همراه پایین­ترین میزان کالوس­زایی و شیشه­ای شدن را نشان داد. بیشترین درصد کالوس­زایی (98/60% و 11/61%) در دوره­های زمانی بالاتر (180 و 240 ثانیه) امواج فراصوت مشاهده گردید. بیشترین درصد ساقه­های چندگانه، تعداد ساقه در هر ریز نمونه و درصد ریز نمونه­های شیشه­ای شده در تیمار 30 ثانیه امواج فراصوت بدست آمد. افزایش دوره­های زمانی تیمار با امواج فراصوت باعث کاهش میزان زنده­مانی سلول‌ها و بافت‌های گیاهی و در نتیجه کاهش ساقه­دهی ریزنمونه­ها گردید.
