بیماری زنگ ساقه گندم با عامل قارچی Puccinia graminis f. sp. tritici یکی از مهم­ترین بیماری ­های این محصول در جهان می ­باشد. یافتن منابع مقاومت جدید نسبت به این بیماری در ژنوتیپ ­های سینتتیک گندم هگزاپلویید به ­دلیل داشتن تنوع ژنتیکی بالا محتمل می­ باشد. ﺑﻪ­ﻣﻨﻈﻮر آگاهی از ساختار ژنتیکی جمعیت عامل بیماری زنگ ساقه، 12 جدایه تک جوش شده مربوط به مناطق گرگان و بروجرد در سال­ های 1393 و 1394 تعیین نژاد شدند، سپس مقاومت 348 ژنوتیپ سینتتیک گندم با استفاده از صفت تیپ آلودگی در مقیاس صفر تا 4 نسبت به چهار جدایه از 12 جدایه تعیین نژاد شده در مرحله گیاهچ ه­ای با استفاده از طرح بلوک ­های کامل تصادفی در دو تکرار مورد بررسی قرار گرفت. بر اساس نتایج حاصل از واکنش ارقام استاندارد و افتراقی گندم ضمن شناسایی دو نژاد TKTTF و TTTTF در بین تمامی جدایه­ ها، بیماری­زایی چهار جدایه­ ی منتخب بر روی ژنوتیپ­ های حامل ژن­ های Sr5، Sr6، Sr7a، Sr7b، Sr8a، Sr8b، Sr9a، Sr9b، Sr9d، Sr9e، Sr9g، Sr10، Sr12، Sr14، Sr15، Sr16، Sr17،Sr18 ، Sr19، Sr20، Sr21، Sr22، Sr23، Sr28، Sr30، Sr34، Sr36، Sr37، Sr38، SrTmp و SrMcN مشاهده گردید. تجزیه واریانس مرکب نشان داد که در مرحله­ی گیاهچ ه­ای بین ژنوتیپ­ های گندم از نظر واکنش به جدایه­ های منتخب اختلاف معنی ­داری داشتند. همچنین معنی­ دار شدن اثر متقابل جدایه × ژنوتیپ حاکی از آن بود که در برخی از ژنوتیپ­ ها مقاومت اختصاص به­ جدایه داشتند. واکنش اختصاص به جدایه در 16 درصد از ژنوتیپ ­ها مشاهده گردید، بدین معنی که حداقل در برابر یکی از جدایه ­ها واکنش مقاومت و در برابر جدایه ­های دیگر حساسیت وجود دارد. 61 درصد ژنوتیپ­ ها نسبت به هر چهار جدایه حساس (تیپ آلودگی 3 تا 4) و 23 درصد ژنوتیپ­ ها (مثل لاین­ های 22842، 22857، 22862، 22863، 22864) نسبت به همه­ ی آن­ها مقاوم بودند (تیپ آلودگی صفر تا 2)، ژنوتیپ­ های مقاوم دارای ژن­ های مقاومت موثر گیاهچ ه­ای­ بوده و می­ توانند در برنامه ­های به ­نژادی مقاومت به زنگ ساقه مورد استفاده قرار گیرند. نتایج این تحقیق اولین گزارش از مقاومت ژنوتیپ­ های سینتتیک گندم نسبت به نژادهای غالب زنگ ساقه در ایران می­ باشد.
