در این مطالعه، به‌منظور تعیین مناسب‌ترین روش القای پلی‌پلوئیدی در گیاه خشخاش برگ‌ریزان آزمایشی به صورت فاکتوریل در قالب طرح کاملاً تصادفی با سه تکرار و سه فاکتور شامل سطوح مختلف کلشی‌سین (0، 05/0، 1/0، 2/0 و 5/0 درصد)، مدت زمان تیمار (12، 24 و 48 ساعت) و نوع ریزنمونه (بذر، گیاهچه و جوانه انتهایی ساقه)، در سال‌های 1397-1396 در دانشگاه محقق اردبیلی انجام شد. در کنار ارزیابی‌های مورفولوژیکی و روزنه‌ای، روش فلوسایتومتری افزایش محتوای ماده ژنتیکی را در برخی از گیاهان تیمار شده تأیید نمود. نتایج نشان داد که تیمار جوانه انتهایی ساقه با غلظت کلشی‌سین 05/0 یا 1/0 درصد در زمان تیمار‌ 48 ساعت، به دلیل دارا بودن بیشترین درصد زنده‌مانی (%4/44) و القای تتراپلوئیدی (%9/18) بهترین روش تولید گیاهان تتراپلوئید در خشخاش برگ‌ریزان است. با بررسی اثرات اتوپلی‌پلوئیدی بر برخی صفات مورفوفیزیولوژیک مشخص شد که میانگین فواصل میانگره، ارتفاع بوته و تراکم سلول‌های روزنه در گیاهان تترا‌پلوئید کاهش و اندازه و ضخامت برگ، اندازه کپسول، تعداد برگ، وزن خشک کل بوته، اندازه سلول‌های روزنه‌ای، محتوای کلروفیل، پروتئین، کربوهیدرات و فعالیت آنزیم‌های کاتالاز و پراکسیداز افزایش یافته است. این در حالی است که تغییری در میزان فعالیت آنزیم پلی‌فنل اکسیداز در نتیجه القای تترا‌پلوئیدی مشاهده نشد. علاوه بر این، افزایش محتوای مورفین و کاهش محتوای کدئین در گیاهان تترا‌پلوئید حاصل مشاهده شد. در مجموع با توجه به اینکه کلشی‌سین نقش مؤثری در القای تتراپلوئیدی گیاه خشخاش برگ ریزان و تغییر در خصوصیات مورفولوژیکی و فرآیندهای متابولیکی آن دارد میتوان از این نتایج در برنامه‌های به‌نژادی این گیاه دارویی استفاده کرد.
